  • - Methyl gallate và quercitrin có tương quan chặt chẽ với tổng các hợp chất phenol và với tổng chất chống oxy hóa, vì vậy có thể dựa vào hàm lượng methyl gallate và quercitrin để đánh giá nhanh khả năng tổng các hợp chất phenol cũng như tổng chất chống oxy hóa của 7 loài dược liệu.

  • KẾT LUẬN

  • 1. Hoạt tính chống oxy hóa của các cao chiết từ 7 loài dược liệu

  • 1.1. Cao toàn phần của 7 loài dược liệu được nghiên cứu đều thể hiện hoạt tính chống oxy hóa rất tốt trong mô hình thử nghiệm bắt gốc tự do DPPH với các giá trị IC50 dao động từ 1,20 ÷ 17,53 µg/mL, mạnh gấp khoảng 2 ÷ 32 lần curcumin với IC50 là 38,50 µg/mL. Đặc biệt, Gối hạc có hoạt tính mạnh gấp 32 lần curcumin. Tuy nhiên, trong mô hình thử nghiệm cho electron với molipdenum, chỉ có cao toàn phần của Mán đỉa ở nồng độ thấp (0,1 mg/mL) có lực chống oxy hóa tương đương curcumin ở cùng nồng độ, nhưng xét về hàm lượng thì tổng chất chống oxy hóa trong Chùm gởi lại nhiều nhất.

  • 1.2. Tương tự cao toàn phần, tất cả các cao phân đoạn từ 7 loài dược liệu đều thể hiện hoạt tính chống oxy hóa cao hơn curcumin trong mô hình cho nguyên tử hydro, trừ cao n-hexane của Chùm gởi, cao n-butanol của Chanh ốc và cao n-hexane của Rạng đông thấp hơn curcumin một ít. Đáng chú ý, cả 5 cao phân đoạn của cây Cổ ướm cũng thể hiện hoạt tính chống oxy hóa cao hơn cucurmin trong cả hai mô hình bắt gốc DPPH và mô hình cho electron với molipdenum. Mỗi loài dược liệu trong 6 loài còn lại đều có từ 1 đến 2 cao phân đoạn có hoạt tính bắt gốc DPPH rất tốt với IC50 ở trong khoảng từ 1/2 đến 1/21 so với curcumin.

  • 1.3. Trong các cao phân đoạn, đáng chú ý nhất là cao ethyl acetate của Mán đỉa: có hoạt tính chống oxy hóa - dập tắt gốc DPPH tốt nhất với giá trị IC50 chỉ bằng 1/22 so với curcumin, đồng thời cũng thể hiện lực chống oxy hóa - cho electron tốt nhất. Mặt khác, trong thử nghiệm in vitro trên gan chuột, cao này cho giá trị ED50 thấp (0,63 µg/mL, chỉ bằng khoảng 1/6 ED50 của curcumin là 4,43 µg/mL). Thêm vào đó, trong mô hình thực nghiệm in vivo gây tổn thương gan chuột nhắt trắng bằng paracetamol, cao ethyl acetate từ Mán đỉa ở liều 500 và 1000 mg/kg/ngày thể hiện rõ tác dụng bảo vệ gan, ở liều 2000 mg/kg/ngày có tác dụng bảo vệ gan tương đương với silymarin ở liều 50 mg/kg/ngày.

  • 2.1. Từ phần trên mặt đất của 2 loài Cổ ướm và Mán đỉa cùng thuộc chi Archidendron, đã phân lập và xác định được cấu trúc hóa học của 16 hợp chất. Trong số đó, loài Cổ ướm (A. bauchei) lần đầu tiên được nghiên cứu về thành phần hóa học và từ loài này đã phân lập được 10 hợp chất. Có 8 hợp chất lần đầu tiên được phân lập từ chi Archidendron gồm: lup-20(29)-en-3-one, α-tocospiro A, -spinasterol, -spinasterone, daucosterol, 1-octacosanol, betulinic acid, và α-tocopherol; 8 hợp chất còn lại là: docosenoic acid, stigmasterol, oleanolic acid, methyl gallate, rutin, quercetin, quercitrin và 7-O-galloyltricetiflavan.

  • 2.2. Trong số 16 hợp chất đã phân lập được, 6 hợp chất có hoạt tính bắt gốc DPPH mạnh với IC50 nhỏ hơn nhiều so với curcumin. Quercitrin và methyl gallate được xác nhận chống oxy hóa mạnh trong cả 3 mô hình bắt gốc DPPH, in vitro trên gan chuột và hóa tính toán; hoạt tính của quercertin và rutin được xác nhận trong thử nghiệm DPPH và hóa tính toán; hoạt tính của 7-O-galloyltricetiflavan mạnh trong thử nghiệm bắt gốc DPPH và in vitro trên gan chuột; α-tocopherol (vitamin E) có hoạt tính mạnh trong bắt gốc DPPH. Ở nồng độ 20 µg/mL và 100 µg/mL, kết quả thử nghiệm bắt gốc tự do DPPH và mô hình sinh học chống oxy hóa - bảo vệ gan in vitro trên gan chuột của 6 trong 16 hợp chất trên có mối tương quan chặt chẽ. Kết quả này chứng tỏ các hợp chất trên thể hiện hoạt tính chống oxy hóa - bảo vệ gan chủ yếu theo cơ chế cho nguyên tử hydro. Điều này được xác nhận một lần nữa trên cơ sở đánh giá hoạt tính chống oxy hóa theo phương pháp tính toán năng lượng phân ly liên kết O-H.

  • 3. Hàm lượng và mối tương quan giữa các thành phần có hoạt tính chống oxy hóa

  • Đã xác định hàm lượng của 5 hoạt chất: α-tocopherol, methyl gallate, rutin, quercetin và quercitrin trong 7 loài dược liệu ở Quảng Trị, các loài này có chứa các hoạt chất chống oxy hóa mạnh với lượng tương đương hoặc lớn hơn một số loài dược liệu khác đã được công bố. Hàm lượng tổng các hợp chất phenol trong 7 loài dược liệu dao động từ 16,66 ÷ 93,22 mg GA/g, tổng flavonoid 5,62 ÷ 71,69 mg QU/g, trong đó Cổ ướm và Chùm gởi có hàm lượng cao gấp 3÷4 lần so với 5 loài còn lại. Tổng hàm lượng của 5 hoạt chất có tương quan chặt chẽ với tổng các hợp chất phenol và với tổng chất chống oxy hóa, cho thấy 5 hoạt chất này có sự đóng góp đáng kể vào hàm lượng tổng các hợp chất phenol cũng như hàm lượng tổng chất chống oxy hóa. Trong đó, methyl gallate và quercitrin có tương quan chặt chẽ với tổng các hợp chất phenol và với tổng chất chống oxy hóa, vì vậy có thể dựa vào hàm lượng methyl gallate và quercitrin để đánh giá nhanh khả năng tổng các hợp chất phenol cũng như tổng chất chống oxy hóa của 7 loài dược liệu.
