Introduction Rheumatoid arthritis (RA) is a chronic inflammatory disease characterised by the dysregulated expression of many proinflammatory cytokines including tumour necrosis factor α (TNF-α), with increased yet insufficient production of anti-inflammatory cytokines including IL-10 [1]. The vali- dation of TNF-α as a therapeutic target in RA has encour- aged the investigation of signalling pathways regulating its production by cells relevant to the pathophysiology of this disease. One pathway known to downregulate proinflam- matory TNF-α production and, consequently, upregulate the anti-inflammatory cytokine IL-10 is that elicited by the second messenger cAMP [2,3]. This pathway may there- fore represent a good therapeutic target due to its oppos- ing effects on TNF-α and IL-10. Previously, we and others demonstrated that rolipram, a phosphodiesterase (PDE) IV inhibitor, reduced the clinical and histological severity of collagen-induced arthritis (CIA) [4,5]. Abstract Vasoactive intestinal peptide (VIP) is an anti-inflammatory immunomodulatory neuropeptide with therapeutic potential demonstrated for collagen-induced arthritis. The aim of this study was to characterise its potential anti-arthritic effect on human monocytes, macrophages, T cells, and rheumatoid arthritis synovial membrane cells. Monocytes, macrophages, and T cells derived from human peripheral blood were treated with VIP and compared with other cAMP-elevating drugs for a range of activating stimuli. Cytokine production was assessed for cell cultures and, in addition, the ability of VIPs to activate cAMP response element binding protein. VIP partially suppressed monocyte- and macrophage-derived tumour necrosis factor α (TNF-α) with no effect on IL-10, whereas VIP fails to regulate IL-10 and TNF-α production by T lymphocytes. No such modulation of cytokine profile was observed for rheumatoid arthritis synovial membrane cells. Elevation of intracellular cAMP, on the other hand, potently suppressed macrophage TNF-α production and modulated T- cell response by inhibiting TNF-α and IFN-γ. Pursuant to the Government's Decree No 63/2014/NĐ-CP dated June 26, 2014 on guidelines for some Articles on contractor selection of the Law on Bidding; Pursuant to the Government's Decree No 116/2008/NĐ-CP dated November 14, 2008 on defining the functions, tasks, powers and organizational structure of the Ministry of Planning and Investment; The Minister of Planning and Investment promulgates a Circular to provide guidelines for planning contractor selection Article Scope of adjustment This Circular provides specific guidance on making, submitting, appraising, approving contractor selection plans with regard to the procurements and projects regulated by the Law on Bidding No 43/2013/QH13, except for drug procurement prescribed in Clause Article of the Law on Bidding Article Subject of application This Circular applies to agencies, organizations participating in making, submitting, appraising, approving contractor selection plans with regard to the procurements and projects mentioned in Article of this Circular, and relevant entities Article The application of this Circular In case an international agreement contains regulations on contractor selection procedures, Clause and Clause Article of the Law on Bidding shall apply Procedures for submitting, appraising, and approving the contractor selection plan shall comply with this Circular In case an international agreement does not contain regulations on contractor selection procedures, the contractor selection plan shall be made, submitted, appraised, and approved in accordance with this Circular For procurements executed before a decision on project approval, the following adjustments must be made when applying this Circular: a) If the investor is determined, the phrase “investor” will be replaced with “Units directly under investor”; “Decision to approve the project” with “Relevant Decisions from unit’s leaders appointed to prepare the project” With regard to procurements funded by state budget prescribed in Point d Clause Article of the Law on Bidding, this Circular must be applied when making, submitting, appraising, and approving contractor selection plans, except for the contents prescribed by the Circular on use of state budget for regular operation promulgated by the Ministry of Finance When applying this Circular, the phrase “Investor” will be replaced with “procuring entity”; “project” with “procuring estimation” Article Application for contractor selection plan approval The rules for making the contractor selection plan are specified in Article 33 of the Law on Bidding The basis for making the contractor selection plan is specified in Article 34 of the Law on Bidding The application for contractor selection plan approval is made according to Form enclosed herewith, which contains: a) Completed tasks including tasks related to preparation of project and procurements to be completed first with corresponding values and legal basis; b) Tasks that not apply any contractor selection method include: operation of project management board, provision of compensation for land clearance, loan interest payment, tasks performed by the investor including making, appraising the request for expression of interest, pre-qualification documents, bidding documents, request for proposals; assessing expressions of interest, prequalification applications , bid-envelopes, proposals; appraising the contractor selection result, and other tasks that not apply contractor selection with corresponding values; As 85% of PC express LHRH receptors, we investigated the interaction of goserelin incubation with radiotherapy under androgen-deprived conditions in vitro. Methods: LNCaP and PC-3 cells were stained for LHRH receptors. Downstream the LHRH receptor, changes in protein expression of c-fos, phosphorylated p38 and phosphorylated ERK1/2 were analyzed by means of Western blotting after incubation with goserelin and irradiation with 4 Gy. Both cell lines were incubated with different concentrations of goserelin in hormone-free medium. 12 h later cells were irradiated (0 – 4 Gy) and after 12 h goserelin was withdrawn. Endpoints were clonogenic survival and cell viability (12 h, 36 h and 60 h after irradiation). Results: Both tested cell lines expressed LHRH-receptors. Changes in protein expression demonstrated the functional activity of goserelin in the tested cell lines. Neither in LNCaP nor in PC-3 any significant effects of additional goserelin incubation on clonogenic survival or cell viability for all tested concentrations in comparison to radiation alone were seen. Conclusion: The clinically observed increase in tumor control after combination of goserelin with radiotherapy in PC cannot be attributed to an increase in radiosensitivity of PC cells by goserelin in vitro.
