thứ 21 đứng trước nhiều vấn đề cần phải đối mặt Trong vấn đề thiếu lượng vấn đề cấp bách mang tính toàn cầu cần phải sớm có biện pháp giải Những nguồn lượng hóa thạch than đá, dầu mỏ, khí thiên nhiên vô tận Với phát triển ngành công nghiệp giới hiện nguồn lượng nhanh chóng cạn kiệt gây ảnh hưởng đến phát triển xã hội đời sống người Hơn nữa, đặc điểm tài nguyên hóa thạch thành phần hóa học có chứa nguyên tố cacbon, hydro, oxy, nitơ, lưu huznh, đặc biệt than có kèm theo chất phóng xạ urani thori, sử dụng nguồn nhiên liệu gây ảnh hưởng lớn môi trường *1+ Trong việc khai thác nhiên liệu sinh học phương thức tạo lượng quan tâm hiện hiệu suất cao, tốn thân thiện với môi trường *4+ Điển hình việc khai thác nguồn lượng hydro sinh học - nguồn lượng thay vô cùng ưu tương lai từ chủng vi sinh vật kị khí ưa nhiệt cao Thermotoga neapolitana DSM 4359[20] Đây hướng mẻ đầy triển vọng không phần khó khăn nhiều thách thức Do tiến hành thực hiện đề tài: “Nghiên cứu trình sản xuất hydro sinh học từ rác thải nông nghiệp nhờ chủng vi khuẩn ưa nhiệt Thermotoga neapolitana DSM 4359” với mục tiêu sau: Xây dựng quy trình sản xuất hydro sinh học từ số chất khác (glycerol, xylose, glucose…) Đưa kết thăm dò số nguồn phế thải nông nghiệp làm nguyên liệu đầu vào cho trình sản xuất hydro sinh học nhờ lên men vi khuẩn kị khí ưa nhiệt Thermotoga neapolitana DSM 4359 CHƢƠNG - TỔNG QUAN TÀI LIỆU 1.1 Thực trạng nguồn lƣợng giới nhu cầu tìm nguồn lƣợng thay 1.1.1 Dầu mỏ Nguồn tài nguyên thiên nhiên từ biển trở thành tiêu điểm cạnh tranh quốc tế Dầu mỏ coi nguồn lượng cho toàn giới tới năm 2025 1.1.2 Khí tự nhiên Cùng với dầu mỏ, gần đây, khí thiên nhiên coi nguồn nhiên liệu có nhu cầu tiêu thụ lớn giới với nhu cầu hàng năm tăng nhanh 1.1.3 Than Là nguồn nhiên liệu hóa thạch sử dụng từ lâu giới Tổng trữ lượng than toàn giới ước tính khoảng 1.083 tỷ tấn, đủ cung cấp cho khoảng 170 năm với mức tiêu thụ hiện 1.2 Hydro nguồn lƣợng ƣu việt Hydro nguyên tố hóa học hệ thống tuần hoàn nguyên tố với nguyên tử khối Ở trạng thái tự điều kiện bình thường, hydro không màu, không mùi không vị, tỉ trọng 1/14 tỉ trọng o không khí Nhiệt độ cháy hydro lên đến 3.000 C cháy ôxy, cao so với tất loại khí o o khác khí methane (CH4) đạt 2.148 C, propane (C3H8) đạt 2.385 C [32, 34] 1.3 Một số phƣơng pháp sản xuất hydro sinh hoc 1.3.1 Sản xuất hydro điện Sản xuất H2 điện cách cho dòng điện qua điện dẫn điện nước (kiềm polyme), kết tách phân tử H2O thành H2 oxy (O2) Hydro sản xuất điện có chất lượng tương đối cao, carbon, lưu huznh, hợp chất tạo trình 1.3.2 Sản xuất hydro khí metan (SMR) Quá trình SMR thường sử dụng nước nhiệt độ cao (800oC) phản ứng với khí mêtan (CH4), tạo thành H2 carbonmonoxide (CO) Sau khí CO phản ứng với nước để tạo thành carbondioxide (CO2) bổ sung H2 1.3.3 Sản xuất Hydro nước oxy hóa khuẩn kị khí không bắt buộc 1.5.3 Vi khuẩn hiếu khí 1.6 Giới thiệu Thermotoga neapolitana 1.6.1 Lịch sử Thermotoga neapolitana phân lập lần đầu vào năm 1986, trầm tích núi lửa biển nông gần Lucrino, vịnh Naples, Italia *35+ 1.6.2 Phân loại Thermotoga neapolitana thuộc: Siêu giới: Vi khuẩn; Ngành: Thermotogae; Lớp: Thermotogae; Bộ: Thermotogale; Họ: Thermotogaceae; Chi: Thermotoga [33] 1.6.3.Đặc điểm hình thái sinh thái • Tế bào hình que, Gram âm, tồn riêng biệt theo cặp; dài: 1,1 - 1,5 μm, rộng: 0,6 μm 1.6.4 Đặc điểm hệ gen • NST dạng tròn • Hệ gen có 1800 kb • Tỉ lệ G + C 41,3 % *35+ Đặc sắc kí khí GC - Hệ thống sắc kí lỏng cao áp - Nồi lên men 5L 2.2.2 Các dụng cụ Bình serum 30ml, nắp cao su nắp nhôm, kim tiêm để nuôi cấy kị khí Gas-tight syringe (P/N 008160; 1MRVLL-GT; C04-A1831- Australia) để lấy mẫu khí cho GC, dụng cụ kẹp nắp nhôm Handy crimper for aluminum seal 2.3 Môi trƣờng nuôi cấy 2.3.1 Môi trường nuôi cấy với nguồn chất đơn giản 2.3.2 Môi trường nuôi cấy với nguồn chất bã đậu Tiền xử lí bã đậu: • Bã đậu mang xử lí nhiệt 90o 15 phút để tiêu diệt vi khuẩn tiêu thụ Hydro • Các chỉ tiêu COD , tổ ng Nitơ, tổ ng Photpho đươ ̣c phân tích ta ̣i Viê ̣n Hoá bản - Trường Đa ̣i ho ̣c Bách khoa Hà Nội 2.3.3 Môi trường nuôi cấy với nguồn chất glycerol 2.3.4 Môi trường nuôi cấy với chất rơm, rạ 2.4 Phương pháp sục khí nitơ và kiểm soát pH làm tăng hiệu suất tạo thành bào đánh giá khối lượng tế bào khô (dry cell weight - DCW) mật độ tế bào (opical density - OD600) đo máy đo OD Khí H2 headspace lấy gas-tight syringe, thể tích lần hút 100 µL (Australia) xác định phương pháp sắc kí khí GC Acid acetic acid lactic xác định hệ thống sắc kí lỏng cao áp Kế t quả thu đươ ̣c đươ ̣c xử lý bằ ng phương pháp thố ng kê sinh ho ̣c CHƯƠNG - KẾT QUẢ VÀ THẢO LUẬN 3.1 Nghiên cứu tạo thành hydro chủng Thermotoga neapolitana DSM 4359 từ nguồn đường đơn 3.1.1 Hoạt hóa, nuôi cấy chủng Thermotoga neapolitana DSM 4359 Dựa vào kết đo OD đo lượng khí hydro máy GC, nhận thấy chủng vi khuẩn Thermotoga neapolitana DSM 4359 sinh trưởng tốt sinh khí hydro 3.1.2 Ảnh hưởng nồng độ chất đơn giản đến sản sinh hydro chủng Thermotoga neapolitana DSM 4359 Có thể thấy: hàm lượng hydro cao thu nồng độ chất glucose, xylose 5g/L, với hàm lượng là: 19.68 20.01(mmol/L) 3.1.3 Khảo sát số nguồn nitơ phổ biến cho sản xuất hydro sinh học từ chủng vi khuẩn kỵ khí ưa nhiệt DSM 4359 đánh giá khả sinh hydro Do đó, thấy hàm lượng hydro cao thu nồng độ nguồn nitơ cao nấm men, peptone NH4 NO3 2g/L, với hàm lượng là: 20.01; 19.27; 13.16 (mmol/L) 3.2 Nghiên cứu ảnh hƣởng nhiệt độ tới trình sản xuất hydro DSM 4359 o Khi nhiệt độ nuôi cấy tăng từ 50 đến 75 C lượng hydro sinh phát triển tế bào tăng dần đạt o giá trị cực đại 75 C, với sản lượng hydro thu tăng từ 9.82 đến 20.09 (mmol/L) giá trị OD tăng từ 0.15 đến 0.39 3.3 Nghiên cứu ảnh hưởng pH tới trình sản xuất hydro chủng vi khuẩn Thermotoga neapolitana DSM 4359 Khi giá trị pH tăng từ đến 7.5 cho thấy có tăng tích lũy hydro nhanh chóng từ 6.56 đến 20.13 mmol/L Sự phát triển vi khuẩn đạt giá trị cực đại OD600 = 0.39 giá trị pH 3.4 Nghiên cứu phƣơng pháp sục khí nitơ làm giảm áp suất khí bình nuôi cấy để tăng hàm lƣợng hydro Hơn nữa, điều kiện sục khí nitơ kiểm soát pH, sản xuất hydro cao nhiều so với sục khí nitơ không bao gồm kiểm soát pH Lượng khí hydro thu áp dụng phương pháp sục khí nitơ kiểm soát pH tăng lên đến 2.98±0.1 mol hydro/mol glucose tiêu thụ sau nuôi cấy 3.5 Hàm lƣợng acid hữu đƣợc tạo trình sản xuất hydro T neapolitana Sau 24h nuôi cấy, pH môi trường giảm nhanh xuống mức tối ưu (7-6.8), cùng với đó, tốc độ sinh hydro giảm dần Các acid hữu acid lactic acid acetic tạo liên tục song song với việc tạo hydro tốc độ sản xuất chúng tăng nhanh từ 12 đến 36h nuôi cấy 3.6 Nghiên cứu ảnh hƣởng chất vô tới trình sản xuất hydro T neapolitana Vậy nồng độ tối ưu KH2PO4, MgCl2.6H2O, Na2HPO4.12H2O, NaCl 150mg/L, 20mg/L, 400mg/L 20g/L, cách riêng rẽ 3.7 Bước đầu thăm dò, nghiên cứu tạo thành hydro từ nguồn rác thải nông nghiệp và nguồn phụ phẩm trình sản xuất bio-diezel nhờ lên men kị khí chủng vi khuẩn DSM 4359 3.7.1 Khảo sát lựa chọn loại nguyên liệu đầu vào-phụ phẩm trình sản xuất bio-diezel cho tạo thành hydro Hiệu suất hydro tạo sau 49h glycerol tinh khiết phụ phẩm glycerol 1.02 ± 0.05 mol H2/mol glycerol tiêu thụ 1.28± 0.06 mol H2/mol glycerol tiêu thụ 3.7.2 Xác định thành phần dinh dưỡng khác cần bổ sung cho trình sinh trưởng trình tạo thành H Khảo sát lựa chọn loại nguyên liệu đầu vào - phụ phẩm trình sản xuất sản xuất đậu phụ Với chất bã đậu thô, sản lượng hydro thu tương đối thấp, đạt giá trị cao 5.2 mmol/L thời điểm 42h đến 48h Giá trị pH giảm dần từ 7.5 đến 5.7 sau 48h nuôi cấy Khi sử dụng bã đậu qua xử l{ làm chất cho thấy sản lượng hydro cao nhiều so với lượng hydro sử dụng bã đậu thô làm chất Lượng hydro thu tăng lên khoảng 2.8 lần đạt giá trị cao 14.6 mmol/L thời điểm 48h nuôi cấy 3.7.3 Khảo sát lựa chọn loại nguyên liệu đầu vào chế phẩm phân hủy lignocellulose từ rơm rạ cho tạo thành hydro 3.7.3.1 Nghiên cứu điều kiện thích hợp cho tiền xử lý rơm rạ o o o Ở nhiệt độ trung bình từ 80 C đến 100 C/30 phút loại bỏ 3,0 - 7,0% lignin Ở nhiệt độ 121 C loại bỏ đượ c tỷ lệ lignin cao nhât́ là 55,79% o Tăng thời gian tiền xử lý bằng dung di ̣ch ammoniac 10% từ 10 phút đến 60 phút 121 C thu kết 60 phút loại bỏ đượ c 65,21 % lignin Hàm lượng glucose xử l{ 57,07 % o Tiền xử lý vớ i axit sulfuric 1% nhiệt độ 121 C thờ i gian 50 phút cho hiệu quả cao nhât́ xử l{ đượ c 65,97% xylose 3.7.3.2 Thành phần rơm rạ sau tiền xử lý Theo phương pháp 2.3.4, có kết phân tích thành phần rơm rạ sau: 41% cellulose; 17.8% lignin; số chất khác 3.7.3.3 Sản lượng hydro sử dụng chất rơm rạ qua xử lý Để xác định ảnh hưởng chất rơm rạ đến sản lượng hydro, chủng nghiên cứu nuôi cấy thời gian từ 24 đến 120 sử dụng chất rơm rạ thô rơm rạ qua tiền xử l{ với nồng độ 5g/L tiền xử l{ rơm rạ giúp làm tăng sản lượng hydro cách đáng kể so với không qua tiền xử l{ Sản lượng hydro đạt mức cao nhất11.52 mmol/L, cao gần lần so với sử dụng rơm rạ thô làm chất (3.28 mmol/L), với 85.4% chất tiêu thụ, mức độ chuyển hóa đường 95.7% xylose 73% với glucose 3.8 Nghiên cứu trình sản xuất hydro phương pháp lên men theo mẻ có bổ sung (fed-batch culture) sử dụng nồi lên men L Để sản xuất hydro liên tục làm tăng sản lượng hydro Phương pháp lên men theo mẻ có bổ sung sử dụng nồi lên men 5L thực hiện sử dụng hydro trung bình đạt 3.15 mmol/L/h 3.9 Đánh giá khả sử dụng nguyên liệu đầu vào trình sản sinh hydro chủng vi khuẩn nghiên cứuTrên quy mô nhỏ, tiến hành thí nghiệm sử dụng bình với thể tích 120mL Với pH tối ưu ban đầu đưa vào 7.5 nhiệt độ trì mức 70°C KẾT LUẬN Đã hoạt hóa nuôi cấy chủng Thermotoga neapolitana DSM 4359 - chủng vi khuẩn kí khí ưa nhiệt dùng để nghiên cứu sản xuất hydro sinh học Đã xác định nồng độ chất ban đầu phù hợp Đã nghiên cứu khảo sát nguồn nitơ phù hợp 2g/L cao nấm men, cho sản lượng hydro đạt 20.01 mmol/L Đã xác định điều kiện nuôi cấy tối ưu cho chủng Thermotoga neapolitana DSM 4359 nhiệt độ 75°C, pH7.5, tốc độ lắc 100rmp Đã bước đầu tìm hiểu số nguồn chất tái tạo, sẵn có bã đậu, phụ phẩm glycerol từ trình sản xuất dầu diezel sinh học, từ rơm, rạ Hàm lượng hydro đạt là: 15mmol/L; 20mmol/L; 11.5mmol/L, từ nguồn tương ứng Đã nghiên cứu sản xuất hydro quy mô 5L phòng thí nghiệm nghiên cứu trình sản xuất hydro phương pháp nuôi cấy theo mẻ có bổ sung (fed-batch), hiệu suất thu trung bình 3.15 mmol hydro/mol chất KIẾN NGHỊ Tiếp tục sâu tìm hiểu điều kiện nuôi cấy chủng Thermotoga neapolitana DSM 4359 để tối ưu hóa môi trường nuôi cấy nhằm thu nguồn hydro lớn Phát triển quy mô nuôi cấy từ nuôi cấy mẻ sang nuôi cấy liên tục thời gian dài để tìm hiểu khả sản sinh hydro có ứng dụng thiết thực quy mô công nghiệp INTRODUCTION World in the early years
