MỞ ĐẦU Nghiên cứu, tìm kiếm các chất mới có hoạt tính sinh học có sử dụng các hợp chất thiên nhiên làm chất dẫn đường nhằm thiết kế, tổng hợp các chất có thể sử dụng làm thuốc hay trong thiết kế thuốc luôn là một mục tiêu quan trọng của các nhà khoa học. Trong hóa học các hợp chất thiên nhiên và hóa dược, các hợp chất ketone α,βkhông no được phát hiện rất sớm và dường như có mặt trong hầu hết các lớp chất quan trọng của thiên nhiên như alkaloid, terpene, sesquiterpen, triterpenoid, chalcone và flavone như daphnipaxianines trong cây Daphliphyllum paxianum, myrtenal từ loài Citrus reticulata, zerumbone của Zingiber zerumbet, licorisoflavane A, quercetin, kaemferol trong cây Morus alba L. hay isoliquirigenin từ cây Glycyrrhiza glabra và đặc biệt là curcumin từ loài Curcuma longa L., ngay cả chuỗi DNA của cơ thể sống cũng được tạo thành từ các hợp chất chứa nhóm ketone α,βkhông no là thymine và uracil. Tuy vậy, trong tất cả các lớp chất thiên nhiên đã được biết đến thì nhóm ketone α,βkhông no này lại chính là một phần cấu trúc đặc trưng nhất, không thể thiếu trong bất kỳ hợp chất chalcone và flavone nào. Do nhóm ketone α,βkhông no hiện diện trong phân tử nên các hợp chất này có vai trò quan trọng trong cả hóa học và sinh học. Về mặt hóa học, các hợp chất ketone α,βkhông no là chất trung gian chìa khóa để tổng hợp nhiều chất quan trọng như flavonoid, pyrazoline, diazepine, pyrimidine,… Về mặt sinh học, nhóm ketone α,βkhông no được xác định là trung tâm của nhiều hoạt tính sinh học bao gồm hoạt tính kháng viêm, hoạt tính chống sốt rét, hoạt tính chống ký sinh trùng, hoạt tính chống huyết áp hay loại bỏ yếu tố NF-κB gây ra nhiều bệnh khác nhau, đặc biệt là hoạt tính gây độc tế bào do nhóm này được xem như là các Michael acceptor đối với nhóm thiol của một số protein xác định hay khả năng định hướng các tế bào ung thư chết theo lập trình. Chính vì vậy, các hợp chất chứa nhóm ketone α,βkhông no luôn là đối tượng nghiên cứu hấp dẫn của các nhà khoa học, một số thuốc chứa nhóm này cũng đã được sử dụng hiệu quả trong điều trị bệnh như AZT, Edoxudine, Zalcitabine, Griseofulvin và nhiều các chất khác chứa nhóm này phân lập từ thiên nhiên được sử dụng trong hỗ trợ điều trị điều trị bệnh ung thư. Với mục đích đóng góp thêm những nghiên cứu mới về đối tượng này, luận án “Tổng hợp và nghiên cứu hoạt tính sinh học của một số ketone α,βkhông no có cấu trúc tương tự trong thiên nhiên” trình bày một số chuyển hóa mới của zerumbone, tổng hợp một số chacone mới có chứa các nhóm thế khác nhau dựa trên chalcone mẹ có hoạt tính xuất hiện nhiều trong thực vật và sàng lọc hoạt tính gây độc tế bào đối với một số dòng tế bào ung thư người nhằm định hướng cho việc khai thác một số các hợp chất mới có hoạt tính tiềm năng trong lĩnh vực chăm sóc sức khỏe cộng đồng.
